Frampton (25-1, 14 KOs), of Belfast, was the sharper fighter from the opening bell and imposed his will on the veteran Donaire (38-5, 24 KOs) in front of his hometown fans.
Frampton earned a shot at the WBO's regular belt, as well as the IBO featherweight belt, with his triumph.
Fight fans in the United States of America can watch the Frampton vs. Donaire live stream (Video below) the live stream is a free fight streamed via Showtime and YouTube.
The 31-year-old Frampton plans to next fight at Windsor Park, and has been promised the victor of next month's IBF title fight between Lee Selby and Josh Warrington, or WBO champion Oscar Valdez, should he win.
Frampton, who split from promoter Barry McGuigan a year ago, before ditching Shane McGuigan, Barry's son, as his trainer, added: "In the 11th round I was definitely hurt but I survived it and that's what champions do. The only thing on my mind is Windsor Park and I can't wait to get there".
However, if Valdez decides to step up in weight, Frampton would be declared the WBO champion without having to enter the ring. Promoter Frank Warren promotes Frampton, Selby and Warrington.
While the future looks bright for the comebacking Frampton, Donaire appeared to be on the crossroad of a brilliant boxing career.
The loss to Frampton was also his fourth against four of the top featherweight fighters in the world after previously going down against Nicholas Walters, Jessie Magdaleno, and Guillermo Rigondeaux. "As you saw in the 11th round, Nonito Donaire is a unsafe opponent". To his credit, Frampton stepped it up in the sixth and seventh with his jab and left-right combination taking over, yet those uppercuts caused him problems again.
"I think Jamie deserves a lot of credit because the gameplan was flawless", said Frampton. It was the best camp I've ever had and the most scientific approach. Frampton's shots remained crisp and he was in total control, not least in the ninth when the former pound-for-pound contender seemed to turn away, nearly as if it knew he was on his way to only his second defeat in 43 professional fights.
